Recombinase-aided isothermal nucleic acid amplification technology

Recombinase Aided Amplification

RAA technology is short for Recombinase Aided Amplification, a technology that uses recombinase, single-stranded binding protein, and DNA polymerase under isothermal conditions (optimal temperature 39℃) to amplify nucleic acids. The specific principle is: recombinase, single-stranded binding protein, and primers form a polymer that scans double-stranded DNA. At the sequence homologous to the primer, the double-stranded DNA is unwound, the single-stranded binding protein (SSB) binds to the unwound single strand to prevent the renaturation of single-stranded DNA, and in the presence of energy and dNTP, the DNA polymerase completes the chain extension.

Isothermal Amplification Breakthrough: RAA Platform Accelerates Chikungunya Virus Research

Since 2025, Chikungunya virus (CHIKV) has shown exceptionally active spread in tropical regions. Although traditional RT-PCR offers high sensitivity, its >2-hour amplification cycle and heavy reliance on specialized equipment have created bottlenecks for research teams needing critical data during outbreaks. Field traceability studies are further constrained by cold-chain transport requirements and laboratory dependencies.
