Abstract: This study utilized recombinase-mediated isothermal amplification (RAA) technology, selected the I8S rDNA sequence of Plasmodium parasites, and designed universal primers for the detection of Plasmodium. The primers were then screened and their specificity was evaluated. Four primer pairs with particularly good amplification performance were identified. The entire reaction process of this method is carried out at 37°C, with a short amplification time (40 minutes) and excellent specificity. We have successfully established an RAA-based method for detecting Plasmodium parasites, which is well-suited for rapid screening of imported and exported samples at ports of entry and exit.
