Abstract: This study employed recombinase-aided amplification (RAA) technology—specifically, a temperature-controlled method—to target the I8S DNA sequence of Plasmodium parasites. Universal primers for detecting Plasmodium were designed, optimized through rigorous screening, and thoroughly evaluated for their specificity. As a result, four primer sets with superior amplification performance were identified. The entire reaction process was conducted at 37°C, ensuring a rapid amplification time of just 40 minutes while maintaining high specificity. A robust RAA-based assay for Plasmodium detection has been successfully established, making it an ideal tool for the swift screening of imported and exported goods at border checkpoints.
