A one-step reverse transcription recombinase-aided amplification (RT-RAA) assay has been established for the detection of the H7N9 avian influenza virus. Methods: Primers and probes were designed based on the conserved sequences of the HA and NA gene segments of the H7N9 virus, respectively, to develop the RT-RAA method. The specificity and sensitivity of this method were evaluated and compared with those of the Real-time RT-PCR assay. Results: The H7N9 avian influenza virus RT-RAA assay was successfully established, with detection limits of 10 copies/μL for the H7 reaction system and 100 copies/μL for the N9 reaction system. Importantly, no cross-reactivity was observed with other respiratory pathogens. Furthermore, the RT-RAA assay demonstrated high concordance with Real-time RT-PCR, as evidenced by a Kappa coefficient of 0.933. Conclusion: The established RT-RAA assay exhibits rapid, specific, and highly sensitive characteristics, providing a new molecular tool for the swift detection of the H7N9 avian influenza virus.
