A one-step reverse transcription recombinase-mediated isothermal amplification (RT-RAA) assay was developed for the detection of the H7N9 avian influenza virus. Methods: Primers and probes were designed based on the conserved regions of the HA and NA gene segments of the H7N9 avian influenza virus, and an RT-RAA assay was established. The specificity and sensitivity of the assay were evaluated and compared with those of the real-time RT-PCR method. Results: The H7N9 avian influenza virus RT-RAA assay was successfully established. The limit of detection for the H7 reaction system and the N9 reaction system were 10 copies/μL and 100 copies/μL, respectively, and no cross-reactivity was observed with other respiratory pathogens. The results obtained by the H7N9 RT-RAA assay showed high consistency with those from real-time RT-PCR, with a Kappa coefficient of 0.933. Conclusion: The established RT-RAA assay exhibits rapid, specific, and sensitive characteristics, providing a new molecular tool for the rapid detection of the H7N9 avian influenza virus.
