A Fluorescent Recombinase Aided Amplification Assay for Detection of Babesia microt

Abstract: Babesla microl is one of the most common causative agents of babesiosis. A sensitive and rapid detection isnecessary for screening potentialy infected individuals. in this study, B. microti cyto chrome c oxidase subunit l (cox 1) wasselected as the target gene, multiple primers were designed, and optimized by a recombinase-aided amplfication (RAAassay. The optimal primers and probe were labeled with fuorescein. The sensitivity of fuorescent RAA (fRAA) was evalu-ated using gradient dlluents of the cox1 recombinant plasmid and genomic DNA extracted from whole blood of B. microtiinfected mice. The specificity of fRAA was assessed by other transfuslon transmitted parasites. The analytical sensiivityof the fRAA assay was 10 coples of recombinant plasmid per reaction and 10 fg/ul B. microti genomic DNA. No cross-re-action with any other blood-transmited parasites was observed. Our results demonstrated that the fRAA assay would berapid, sensitive, and specific for the detection of B. microti.
