A nucleic acid detection method for Echinococcus granulosus was established, based on recombinase-aided isothermal amplification (RAA) technology. Methods: Specific RAA amplification primers and a fluorescent detection probe targeting a 12S rRNA gene sequence fragment of E. granulosus were designed, screened, and synthesized to develop a fluorescent RAA assay for this parasite. The assay was then evaluated for its detection sensitivity using recombinant plasmids containing varying copy numbers of the target sequence, as well as genomic DNA from E. granulosus at different concentrations. Additionally, the assay’s specificity was assessed by performing fluorescent RAA amplification reactions with genomic DNA samples derived from E. granulosus, Multiceps multilocularis, Schistosoma japonicum, Schistosoma mansoni, Ancylostoma duodenale, Clonorchis sinensis, Taenia saginata, Diphyllobothrium latum, and Taenia solium.
