Based on the FCoV 7b gene sequence, primers and probes were designed, leading to the development of a fluorescent RT-RAA assay for FCoV detection. This method enables rapid pathogen identification within 20 minutes at a constant temperature of 39°C, with no cross-reactivity observed against feline herpesvirus, feline calicivirus, or feline parvovirus. The assay demonstrates a detection limit as low as 1.97 × 10¹ fg/μL for viral nucleic acid. When applied to 30 clinical samples suspected of FCoV infection, the established fluorescent RT-RAA method showed a 93.33% concordance rate when compared to nested PCR. These results confirm that the developed fluorescent RT-RAA assay is highly suitable for the rapid detection of FCoV.
