Development of a Fluorescent RT-RAA Assay for Feline Coronavirus—Zhang Anjie

Based on the 7b gene sequence of feline coronavirus (FCoV), primers and probes were designed, and a fluorescent RT-RAA assay for FCoV was established. This method enables rapid detection of the pathogen within 20 minutes under constant temperature conditions of 39°C, with no cross-reactivity observed against feline herpesvirus, feline calicivirus, or feline parvovirus. The method’s limit of detection for viral nucleic acid is as low as 1.97 × 10¹ fg/μL. Using nested PCR and the established fluorescent RT-RAA assay, 30 clinical samples suspected of FCoV infection were tested, and the concordance rate between the two methods was 93.33%. The results indicate that the developed fluorescent RT-RAA assay for FCoV is suitable for rapid detection of FCoV.
